PCR reagents for linear template generation
- The RTS E. coli Linear Template Set is designed to generate PCR products for use as templates for in vitro protein expression using the RTS 100 E. coli HY Kit. This method of generating DNA templates for protein expression avoids the time-consuming step of cloning into T7-expression plasmids.
To generate transcriptionally active linear DNA fragments, two PCRs must be performed. In the first reaction, genespecific primers are used to add defined overlap regions to a desired gene sequence. The product of the first PCR is mixed with two flanking primers and DNA fragments that code for the T7-regulatory elements and a tag sequence. The first PCR product anneals with the added DNA fragments, and the 3' ends are extended. This final linear-expression construct is further amplified via the flanking primers.
